Total Synthesis of GDGT Lipid Natural Products
The glycerol dibiphytanyl tetraethers (GDGTs) are a class of membrane lipids isolated from prokaryotic organisms known as archaea. These unique lipids possess two glycerol moieties bridged by two isoprenoid-based hydrophobic chains containing up to 8 trans-cyclopentane rings and 16-24 stereogenic centers. The unique structural features of these lipids have inspired a number of groups to study their biophysical properties and biosynthesis. However, individual lipids have not been characterized due to difficulties obtaining the lipids from the producing organisms. Herein, we report a stereoselective synthesis of the archaeal tetraether lipid, parallel GDGT-4.
