Convergent synthesis of the steroidal core of (–)-batrachotoxin
The complex steroidal alkaloid (–)-batrachotoxin (BTX), the primary toxic component in the skin Colombian poison frogs, is a potent allosteric modulator of voltage gated sodium channels (NaVs), the proteins responsible for the generation and propagation of electrical signals in neurons and other excitable cells. Due to its unique ability to influence every measurable aspect of NaV activity, BTX is a promising tool to study these crucial channels. However, chemical synthesis is required to provide the quantities of BTX and derivatives required to drive biological investigations. This work details the development of an efficient, convergent synthesis of the steroidal core of batrachotoxin.
